Effect of Soybean Ethanol Extract (Glycine max.) on NF-kB and Plgf Levels in Huvecs Cultures Induced by Preeclampsia Plasma

Abstract


INTRODUCTION

Preeclampsia is a specific disorder in pregnancy as one of the main causes of neonatal maternal morbidity and mortality. Oxidative stress contributes to the pathomechanism of preeclampsia by increasing transcription factor (NF-kB) and decreasing placental growth factor (PlGF). Antioxidants are used as a therapy to prevent oxidative stress caused by increased free radicals (ROS). This study aims to determine the effect of soybean ethanol extract on NF-kB and PlGF levels in HUVECs cultures exposed to preeclamptic plasma.




METHODS

The study was conducted in vitro using human umbilical vein endothelial cell cultures (HUVECs) and treated with various doses of soybean extract. NF-kB and PlGF levels in each observation group were determined by the enzyme-linked immunosorbent assay (ELISA) procedure. Hypothesis testing was performed with One-Way ANOVA for NF-kB and Kruskal-Wallis for PlGF.




RESULTS

This study found a significant difference (p <0.05) in the mean value of NF-kB between the positive control group and the treatment group given soybean ethanol extract at doses of 70 ppm and 140 ppm. There was also found a significant difference (p <0.05) in the mean value of PlGF between the positive control group and the treatment group that was given soybean ethanol extract at doses of 35 ppm and 70 ppm.




CONCLUSION

Soybean ethanol extract had a significant effect on decreasing NF-kB levels and increasing PlGF levels in HUVECs cultures exposed to preeclamptic plasma. Thus, soybeans containing isoflavones have the potential as a preeclampsia treatment by reducing inflammatory reactions and increasing angiogenesis.
